If you're talking about the video driver, get it from the manufacturer's website. Generally you do not want to download drivers from "Windows Update" if they can be found on the manufacturer's website.
Since this is a desktop PC, I would get them from Nvidia's website.
Before downloading the drivers, you need to make sure you know which video card you have installed. Also make sure you download the 64-bit version, since you have Vista Ultimate x64.
As for Windows Update, make sure you at least download and install all “Important” updates, then look at the “Recommended” files and decide if you want to install or not, (I personally recommend you do), note that drivers can probably be found on the manufacturer’s website as stated earlier.
