How do I configure permissions in Server 2003 to accomplish this..

I have a folder that a number of users have access to. The folder contains
Excel files. I want the users to be able to open and make changes and then
save the Excel file. I do not want users to be able to delete the files nor
do I want the users to create files or folders within this folder.

Is this possible?

Yeah it's possible using NTFS permissions using Group user permissions.
However, you need to post to a NG/forum that has the word Windows in it.
This is a .Net programmer's NG/forum.
