A good approach would be to study for one of the Microsoft DotNet certifications such as Microsoft Certified Solution Developer (MCSD) or Microsoft Certified Application Developer (MCAD). While going through the program would expose you to all the major aspects of the DotNet foundation and its tools and application modes, that alone would not provide you mastery of DotNet. Applying what you learned, over time, might lead to mastery but it depends on your own aptitude and the opportunities you make for yourself.
