Maybe the following article will offer some ideas:

(934562) Error message when you try to update
Windows Defender on a Windows Vista-based
computer: "Error found: Code 0x80240029"
http://support.microsoft.com/kb/934562/en-us

Actually, it fixed nothing. It only replaced the corrupted file/directory.

If you check the logs of Windows Update, you will probably find it had
been finding no updates for a couple of weeks. The problem is that
Windows Update doesn't bother to tell you the same problem is preventing
it from working.

Considering how many lines of code are contained in Vista, you'd think
someone would have added some error handling to notify you of the
problem, advise you to restart Windows and then, execute a script to do
the same thing you did manually.

At least Windows Defender tells you there is a problem. The more
critical Windows Update doesn't bother to do so.
