Sandbox protection

Can someone explain this futher, or send a link to more info..?

If you have UAC and IE protected mode on, how can you still get a malware/virus?

I'd also like to know how to run/test programs in a protected 'sandbox' to make sure it doesn't affect any system files.. VirtualPC?
